What is paraffin microtome?
The paraffin microtome is an auxiliary machine that is widely used in medical and biological research to embed tissue in wax for sectioning. The microtome is a precision mechanical instrument that cuts thin, uniform tissue slices. Usually the case tissue is sealed in paraffin. After solidification, the paraffin is placed at the knife edge of the microtome to cut the paraffin-embedded tissue. Due to the adhesion to the wax edge of the previous slice, a slice strip of multiple slices is made. The tissue sections of the cases made by the paraffin microtome are very thin, the thickness can reach 1-5µm, and the permeability is good. The pathological structure can be carefully observed under the microscope, which can be used to detect various clinical lesions to ensure that doctors can make more accurate judgments, or help researchers to explore at the tissue and cell level.
The paraffin sectioning method includes the steps of sampling, fixing, washing and dehydration, clearing, dipping in wax, embedding, sectioning and sticking, dewaxing, staining, dehydration, clearing, and sealing. Generally, it takes several days from the sampling and fixing of the tissue to the cover slip to make a slide specimen, but the specimen can be stored for a long time and is a permanent microscope slide specimen.
There are five basic types of microtome, which can be divided into:
(1) Shaking microtome Slicer
(2) Rotary microtome slicer
(3) Sliding microtome Slicer
(4) Push type (sled type) microtome slicer
(5) Cryostat.
The most commonly used is the rotary microtome slicer.
